DUKORAL® EFFICACY

Protective efficacy has been evaluated in mass vaccination campaigns and large clinical trials across three continents. Protective efficacy was determined by mathematical modelling. These campaigns and trials have shown that DUKORAL® provides 78–86% protection against cholera in adults.1–5

*The protective efficacy of oral B subunit killed whole-cell (BS-WC) and killed whole-cell (WC) cholera vaccines was assessed in a placebo-controlled, randomised, double-blind study of 63,498 Bangladeshi children aged 2–15 years and women aged over 15 years. Protective efficacy (PE) was estimated as: PE=1-[Proportion of vaccinees ill with cholera]/[Proportion of controls ill with cholera].3
**The PE of oral inactivated whole-cell/recombinant-B-subunit (WC/rBS) cholera vaccine was assessed in a placebo-controlled, randomised, double-blind study of 1,563 Peruvian military recruits. PE was estimated as: PE=([Proportion of controls ill with cholera]-[Proportion of vaccinees ill with cholera])/[Proportion of controls ill with cholera]x100.2
Following a mass-immunisation programme with a two-dose regimen of WC-rBS vaccine, vaccine effectiveness was assessed in 43 persons with cholera and 172 controls during a cholera outbreak in Beira, Mozambique. PE was estimated as: PE=(1-[adjusted odds ratio])x100. Odds ratios were adjusted for: consumption of uncooked food, consumption of dried fish, consumption of food away from home, ownership of multiple kitchen implements, access to a water tap, and residence in a household with electricity.4
Following the receipt of two doses of killed BS-WC cholera vaccine in 23,291 individuals aged two years and older, in six rural and urban sites, efficacy was assessed during an outbreak of cholera. PE was estimated as: PE=(1-adjusted relative risk)x100.5

ADVERSE EVENTS

The most frequently reported adverse reactions relating to DUKORAL® include gastrointestinal symptoms such as abdominal pain, cramps, gurgling, discomfort, diarrhoea and headaches. These adverse events occurred at similar frequency in placebo groups.1
